I immediately plugged in the BAC incinerator and turned it on high. I then plugged in and turned on the vortex. Next, I grabbed medium sized gloves. I made sure that no holes were in my gloves that would potentially expose my hands. I grabbed the 2% bac-down and sprayed in onto my lab station. I used a paper towel to wipe down the station, and through the used paper towel into the orange bag on the floor. After doing this I am now ready to begin.         Dr. Bryns began handing out our single unknown cultures. These samples were brought to the lab as broth. I decided to pick unknown #4. I signed it out and placed my unknown into the test tube rack to keep it from spilling or breaking. I started looking at the flow chart assigned to me by Dr. Bryns that is found in our Supplementary Laboratory Manual (SLM). This chart allowed me to determine my single unknown #4.         I opened by drawer (#9) to retrieve my inoculating loop and needle. I placed both of these into my rack because I was not ready to use them. I then went to the media garage to obtain the properly assigned agar plates. Dr. Bryns had properly laid out the medias with the appropriate dots. I needed 1-Trypticase Soy Agar (TSA) marked with a blue dot, 2-Nutrient Agars (NA) marked with a red dot, and Brain Heart Infusion Agar (BHIA) marked with a purple dot. I went back to my station and began labeling my agar plates. The plates are labeled with the agar at the top, and the lid on the lab table. I started with TSA and labeled the date 02/10/15. I then marked my initials followed by the section number of class DAC-98. I then marked the plate as unknown #4. After that, I wrote TSA to label the agar. Then I wrote 37°C because that is the temperature the plate will be incubated at per Dr. Byrnes. The back of the SLM has the proper labeling technique to label the agar plates. Next, I picked up the first NA plate and labeled the date 02/10/15. I then marked my initials followed by the section number of class DAC-98. I then marked the plate as unknown #4. After that, I wrote NA to label the agar. Then I wrote 37°C because that is the temperature the plate will be incubated at per Dr. Byrnes. Next, I picked up the second NA plate and labeled the date 02/10/15. I then marked my initials followed by the section number of class DAC-98. I then marked the plate as unknown #4. After that, I wrote NA to label the agar. Then I wrote room temperature (RT) because that is the temperature the plate will be incubated at per Dr. Byrnes. Last, I grabbed the BHIA plate and labeled the date 02/10/15. I then marked my initials followed by the section number of class DAC-98. I then marked the plate as unknown #4. After that, I wrote NA to label the agar. Then I wrote CO2 (carbon dioxide) because that is location the plate will be incubated at per Dr. Byrnes. All my medias are now labeled. Finally, I take green tape and label my unknown #4 broth. Using the SLM back cover I label my broth as the following: 02/10/15 DAC-98 single unknown #4 broth. I make sure that the tape creates a tab around the test tube so I can remove it later easily, and so that it doesn’t get tangled with any other unknown broth test tubes.
